Windows 10 64 bit
When I run the daemon tool for open as file I double click auto run.  I get asked to install "Install MechCommander Expansion"

So I click it and get a pop up that says:
Unsupported 16-Bit Application
The program or feature "\??\F:\setup\setup.exe" cannot start or run due to incompatibility with 64-bit versions of Windows.  Please contact software vendor to ask if a 64-bit Windows compatibility version is available.

So what else did I try:
I tried instead of running the auto run to install I clicked on MCX.  It starts up but when I click to launch the first mission I get the same issue as:

The above url I used the information it says for windows 7 even though I have windows 10 and it works.

The basic instructions are:
  • Open the iso as a file using daemon tools.
  • Open a second folder in your program directory (call that folder MCG).
  • Copy all that is in the folder from the iso being launched in daemon tools to the new folder you just named MCG in your program directory.
  • Make a short cut to the MCX app in the MCG folder and place the shortcut on your desktop.
  • Right click on the shortcut and select properties (then select compatibility tab) on your desktop and set these compatibility measures:
After all that it worked for me and hopefully it will work for you.

Don't try to install it on win10. If you have the CD or .ISO image, open that in windows explorer. Copy ALL contents, go to your desktop, create new folder "Mechcommander", open said folder and 'paste'.

Then in your new Mechcommander folder, locate the mechcommander.exe (MCX.exe), right click, copy, paste shortcut on your desktop. Then to run the game, use that shortcut to launch the game. Let me know if you need more help!

Like SeanLang told you, you need to mount a virtual drive with mech commander .ISO file or copy all CD data into a folder you created before in your game directory.
After copying all files just root to the game folder on your computer and start the mech commander *.exe file. With CD the game should start then.

The error you have shown results on the fact that the game exe can not write data in CD drive, with all files copied in a folder on your harddisk it won't appear again.
